Introduction

Paracetamol (also known as acetaminophen) is a widely used analgesic and antipyretic medication. It is commonly used for pain relief and reducing fever.

Conditions of Use (Indications)

  1. Pain Relief: Used to relieve mild to moderate pain, including headaches, toothaches, menstrual cramps, and musculoskeletal pain.
  2. Fever Reduction: Often used to reduce fever in various illnesses, including colds, flu, and infections.
  3. Osteoarthritis: Often used as a first-line treatment for pain management in osteoarthritis due to its effectiveness and safety profile.

Mechanism of Action

Paracetamol is believed to work by inhibiting the cyclooxygenase (COX) enzymes in the brain, which play a key role in the synthesis of prostaglandins. Prostaglandins are chemicals that promote inflammation, pain, and fever. Paracetamol, however, acts predominantly in the central nervous system rather than at the site of inflammation, which differentiates it from nonsteroidal anti-inflammatory drugs (NSAIDs).

Side Effects

  • Rare skin rash or allergic reactions.

2. Serious:

  1. Alcohol: Chronic alcohol consumption can increase the risk of liver toxicity when taking paracetamol.
  2. Warfarin: Regular use of paracetamol may enhance the anticoagulant effects of warfarin, increasing the risk of bleeding.
  3. Rifampin: Rifampin may induce the metabolism of paracetamol, reducing its effectiveness and increasing the risk of hepatotoxicity in overdose.

Contraindications

  1. Severe liver impairment or active liver disease (e.g., cirrhosis).
  2. Hypersensitivity to paracetamol or any component of the formulation.
  3. Chronic alcohol consumption or history of liver disease.
